Methodology: | Fluorescence in situ hybridization (FISH) analysis on interphase nuclei (nuc ish) is performed on paraffin embedded placenta cells using a DNA probe set (AneuVysion: CEP 18/CEP X/CEP Y, LSI 13/21; CEP 16, LSI BCR, Abbott molecular, Inc). |

Recommended Uses: | Stillbirth or spontaneous miscarriages where chromosome analysis is unsuccessful. |
Contraindications: | |
Specimen Requirements: | Paraffin embedded tissue (FFPE): Seven 4-micron sections on immunohistochemistry slides, the middle section stained with H&E |
Collection Kit/Container: | |
Patient Preparation: | |
Collection Instructions: | FFPE tissue: Tissue specimen should be fixed in 10% neutral buffered formalin for 6-48 hours. |
Specimen Handling and Transport: | Store and transport specimens at room temperature (may transport with coolant during hot weather, >85 degrees F). DO NOT FREEZE. The laboratory must receive specimens within 24 hours of collection. |
Unacceptable Conditions: | Necrotic tissue may not contain nuclei with intact DNA. |
Requisition Form: | Cytogenetics Lab Genetic Diagnosis Form #131 |
Required Information: | Laboratory regulations require the following minimum information to be provided on the requisition form for a specimen to be accepted for testing: Patient name or unique identifier; date and time of collection, patient date of birth and sex, specimen type/site of collection, gestational age, test request(s), reason for referral, clinician name and UPIN/NPI, and address for reporting results. Please be certain that name/identifier on the form matches that on the specimen label. |
Results include: | Evidence/no evidence of trisomy 13, 18, and/or 21. Sex chromosome signal pattern. |
Limitations: | FISH results reflect copy number only for the loci of the probes used, and do not rule out isochromosomes or structural abnormalities of chromosomes 13, 18, 21, X, and Y. |
